You can buy a single ISBN, but it's costly. Blocks of 10 run $250 for U.S. individuals/companies; they can be purchased from Bowker's "My Identifiers" site:

Not sure about the reprints question, off the top of my head...
You do not (typically) need to buy the barcode--if you're self-publishing, someplace like LightningSource will do the barcode for you.
But the point of buying your own ISBNs is that you control the info and metadata--that is, you/your company is registered as the publisher, etc. (With someplace like CreateSpace or Lulu, they own the ISBN and the book will always have their metadata attached to it, which marks it immediately as a self-pubbed book--not a problem for everyone, but not everyone knows this.)

One caveat: if you use LightningSource, you really need to know what you're doing; I think CreateSpace and Lulu are more user-friendly for people who've never published/self-published before. LightningSource produces extremely professional results, but they do not offer any real support in building the book (since they're primarily a printer)--I knew this, so I knew I needed to get InDesign and hire a cover designer.
